Clostridium difficile (C. difficile) is a toxin- producing bacterium which is able to colonize the human intestine under given conditions. Released enterotoxins TcdA and TcdB bind to epithelial cells by lectin-glycan interactions and are internalized via endocytosis. As a consequence, toxin-induced inhibition of essential signal transduction pathways causes the destruction of the colon epithelium.

In this project, glycan-functionalized microgels are developed to capture C. difficile toxins TcdA and TcdB. For this purpose, glycans of different length and composition are synthesized enzymatically. The potential of these oligosaccharides to bind the lectin-domain of TcdA and TcdB is tested in an ELISA-type assay.
